BIOAUTOGRAPHY, COMBINATION EFFECTS AND PHOTO-ACTIVATED ENZYMATIC RESTRICTION INHIBITORY ACTIVITY OF ANTIMICROBIAL ALKALOIDS FROM GLYCOSMIS

Glycosmis pentaphylla (Retz.) DC., locally known as nerapan, has long been used in Asian countries a traditional remedy for ailments attributed to microbial infections. This study aims isolate antimicrobial alkaloids from G. pentaphylla, determine their combination effects with selected agents and screen photoactivated enzymatic restriction inhibitory activity. Bioautography-guided isolation of was performed by using column chromatography Staphylococcus aureus, Escherichia coli, Candida albicans the indicator microbes. The combined agents, namely, ciprofloxacin, erythromycin, vancomycin, ketoconazole, were determined checkerboard assay. Photoactivated activity assessed agarose gel electrophoresis. Two active isolated identified arborinine arborine. arborine be range 250 µg/ml 1000 µg/ml. Partial synergy observed all arborine-antibiotics arborinine-ketoconazole interactions against S. aureus C. albicans, respectively. Arborine relatively strongest inhibitor, particularly EcoRI, PstI, SalI. results obtained are promising encourage further research on potential antimicrobial-enhancing agents.

Two new acridone alkaloids from Glycosmis macrantha.
Extraction and chromatographic separation of the extracts of dried stem barks of Glycosmis macrantha lead to isolation of two new acridone alkaloids, macranthanine and 7-hydroxynoracronycine, and a known acridone, atalaphyllidine. The structures of these alkaloids were determined by detailed spectral analysis and also by comparison with reported data.
